The Rams went ahead after 11 minutes when Morgan Whittaker opened his account for the season with a low angled drive.
A brilliant penalty save by Spurs goalkeeper Alfie Whiteman prevented Jahmal Hector-Ingram from doubling the visitors' tally on 29 minutes.
But three minutes later Derby added a second thanks to Duane Holmes's 20-yard shot.
Henrich Ravas tipped a Tariq Hinds effort for Spurs onto the woodwork moments before the home side scored a 69th-minute consolation courtesy of Rodel Richards.
