United went ahead on nine minutes with a stunning Bruno Fernandes volley from Anthony Elanga's cut-back. It was the Portuguese's 50th career goal for the club.
After having a goal overturned following a VAR review, Cristiano Ronaldo scored United's second on the hour, drilling home a penalty after he had been fouled by Rico Henry.
Raphael Varane added a third with a deflected shot 18 minutes from time with his first Premier League goal to seal Man Utd's first win in four top-flight matches and biggest of Ralf Rangnick's reign.
United remain sixth on 58 points, three behind Tottenham Hotspur in fifth spot having played two matches more.
Brentford stay 14th with 40 points, eight clear of the bottom three.
