Wolves started the second half brightly, with Raul Jimenez forcing a save from Paulo Gazzaniga before Adama Traore equalised midway through the second half with a venomous strike.
But late on Vertonghen rose to head in substitute Christian Eriksen's corner to make it four wins from five in the league under head coach Jose Mourinho.
Spurs are now on 26 points, three behind fourth-placed Chelsea and two clear of Wolves in eighth.
