Arsenal went in front on 64 minutes, with Lacazette converting a penalty after he was fouled by Davinson Sanchez.
Spurs were reduced to 10 men in the 76th minute after Lamela received a second yellow card.
Harry Kane then hit a post with a late free-kick and from the rebound, Gabriel headed clear Davinson Sanchez's goalbound effort.
Spurs were unable to find an equaliser as they suffered a first defeat in four matches. They remain seventh on 45 points, with Arsenal still in 10th on 41.
